Background:
Nuclear factor kappa b (NF-kb) is a ubiquitous
transcription factor and an essential mediator of
gene expression during activation of immune
and inflammatory responses. NF-kb mediates
the expression of a great variety of genes in
response to extracellular stimuli including IL-1,
TNFa and bacteria product LPS. NF-kb is
associated with Ikb proteins in the cell
cytoplasm, which inhibit NF-kb activity. The
long-sought Ikb kinase (IKK), which
phosphorylates Ikb, and mediates Ikb
degradation and NF-kB activation, was recently
identified by several laboratories. IKK is a serine
protein kinase, and the IKK complex contains
alpha and beta subunits (IKKa and IKKb). IKKa
and IKKb interact with each other and both are
essential for the NF-kb activation. IKK
phosphorylates both Ikb-a and Ik-. IKK is
expressed in a variety of human tissues.
Specificity:
Binds human IKKb by Western blot. It has no
cross response to IKKa or IKKg.
Positive Control:
Whole cell lysate from Jurkat cells. An 87 kDa
band should be detected by Western blot.
Species Reactivity:
Human, other species not tested.
Ig Isotype:
Rabbit IgG
Immunogen:
A peptide corresponding to amino acids near the
carboxy-terminus of human IKKb (Genbank
accession No. O14920), which differs from the
corresponding murine sequence by one amino
acid.
Format:
Immunoaffinity chromatography purified IgG,
100 μg of polyclonal antibody at 0.5 mg/mL in
PBS containing 0.02 sodium azide.
Storage and Stability:
Stable for at least one year when stored at 4°C.
Applications and Suggested Dilutions:
 Western blot: Use at 1-2 μg/mL.
 Immunocytochemistry: Use at 10 μg/mL.
The optimal dilution for a specific application
should be determined by the researcher.
